Biological Background: HOPX (Homeodomain-only protein) Function and Localization

  • HOPX is an atypical homeodomain protein that lacks DNA-binding activity and functions as a transcriptional repressor.
  • It modulates cardiac growth and development by interacting with serum response factor (SRF), inhibiting SRF-dependent transcription either by blocking DNA binding or recruiting histone deacetylases (HDACs).
  • Overexpression of HOPX can cause cardiac hypertrophy.
  • HOPX may act as a tumor suppressor; it is down-regulated in several cancers including lung cancer, choriocarcinoma, head and neck squamous cell carcinoma, and oral squamous cell carcinoma.
  • It functions as a co-chaperone for HSPA1A and HSPA1B, assisting in chaperone-mediated protein refolding. Related references: PMID:27708256
  • HOPX localizes to both the nucleus and cytoplasm.
  • Alternative splicing produces multiple isoforms, and it contains a characteristic homeobox domain.

Experimental Guidance and Technical Tips

  • The immunogen covers the N-terminal region (aa 1–73) of human HOPX. This region is conserved across species, consistent with the observed cross-reactivity with mouse and rat.
  • For western blotting, expect a band at approximately 8 kDa. Use a low-molecular-weight marker and optimize transfer conditions for small proteins.
  • In immunoprecipitation, pre-clear lysates and include appropriate negative controls to minimize background.
  • For immunohistochemistry (IHC-P) and immunofluorescence (IF-P), validate antigen retrieval and permeabilization protocols in your specific tissue or cell system.
  • The antibody performs well in ELISA applications; consider pairing with a suitable detection reagent.
