Biological Background: HIF1β/ARNT Function and Localization
- HIF1β/ARNT, also known as Class E basic helix-loop-helix protein 2 (bHLHe2) or hypoxia-inducible factor 1-beta (HIF-1-beta), is a transcription factor encoded by the ARNT gene.
- It functions as an obligate heterodimerization partner for several bHLH-PAS proteins, including the aryl hydrocarbon receptor (AHR), mediating xenobiotic response by binding to xenobiotic response elements (XREs) or dioxin response elements (DREs) in target gene promoters. Related references: PMID:28396409, PMID:34521881
- ARNT also heterodimerizes with hypoxia-inducible factor alpha subunits (HIF1A, EPAS1, NPAS1, or NPAS3) to form hypoxia-inducible transcription factors that activate genes involved in cellular responses to hypoxia, including angiogenesis, erythropoiesis, and metabolic adaptation. Related references: PMID:16181639, PMID:20699359
- The protein localizes to the nucleus where it exerts its transcriptional regulatory function.
- ARNT undergoes post-translational modifications such as acetylation, phosphorylation, and ubiquitination (isopeptide bond/ubl conjugation), which may regulate its activity and stability.
- Alternative splicing generates multiple isoforms, contributing to functional diversity.
- Structurally, ARNT contains basic helix-loop-helix (bHLH) and PER-ARNT-SIM (PAS) domains essential for DNA binding and dimerization.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a C-terminal region (aa 465-570) of human HIF1β/ARNT; consider verifying isoform specificity using appropriate controls.
- For IF/ICC, nuclearly localized ARNT may require optimized permeabilization and fixation; include a nuclear counterstain to confirm localization.
- In ELISA, dilute and validate the antibody using known positive and negative samples from human, mouse, or rat to assess cross-reactivity and signal linearity.
- As a polyclonal reagent, anticipate lot-to-lot variation and include reference standards in each experiment.
- Inducing ARNT expression with hypoxia mimetics (e.g., CoCl₂) may enhance detection in cell-based assays.
