Biological Background: HEXA Function and Localization
- HEXA encodes the alpha subunit of beta-hexosaminidase, also known as beta-N-acetylhexosaminidase subunit alpha. It forms the isozymes hexosaminidase A (αβ heterodimer) and S (αα homodimer).
- The enzyme hydrolyzes the non-reducing end N-acetyl-D-hexosamine and sulfated N-acetyl-D-hexosamine from glycoconjugates, including oligosaccharide chains, neutral glycolipids, and mucopolysaccharides.

PMID:16698036
PMID:8123671 - Isozymes S and A are equally active on anionic bis-sulfated glycans, chondroitin-6-sulfate trisaccharide, and dermatan sulfate pentasaccharide, while isozyme B does not hydrolyze these substrates.

PMID:11707436 - Only isozyme A is responsible for the degradation of GM2 gangliosides in the presence of the GM2A activator protein.

PMID:9694901 - The protein is localized to the lysosome and plays critical roles in glycoconjugate and lipid metabolism.
- Mutations in HEXA lead to Tay-Sachs disease, a severe neurodegenerative gangliosidosis characterized by GM2 accumulation.
- Alternative splicing generates multiple transcript variants; the protein undergoes post-translational modifications including glycosylation and disulfide bond formation.
Experimental Guidance and Technical Tips
- The immunogen encompasses amino acids 270–529 of human HEXA, covering a large C-terminal region. This design likely recognizes both full-length HEXA and potential alternative isoforms; verify specificity in your sample system.
- For Western blot, a band near 61 kDa is expected. Use appropriate loading controls and consider reducing conditions to resolve disulfide-linked complexes. Human liver or brain lysates can serve as positive controls.
- For IHC-P and IF-P, standard fixation with 4% PFA and antigen retrieval methods (e.g., citrate buffer, pH 6.0) are suggested. Cross-reactivity with mouse and rat tissues enables preclinical model analysis.
- For ELISA, optimize coating conditions using recombinant HEXA protein. A matched pair with a capture antibody targeting a non-overlapping epitope is recommended for sandwich ELISA development.
