  • HDAC9 is a histone deacetylase that catalyzes the removal of acetyl groups from lysine residues on histones H2A, H2B, H3, and H4, leading to transcriptional repression and epigenetic regulation.
  • It represses MEF2-dependent transcription, playing critical roles in cell cycle progression, developmental events, and skeletal myogenesis inhibition.
  • Isoform 3 lacks catalytic active site residues and is enzymatically inactive; it represses transcription by recruiting HDAC1 and/or HDAC3.
  • HDAC9 protects neurons from apoptosis by inhibiting JUN phosphorylation via MAPK10 and repressing JUN transcription through HDAC1 recruitment.
  • Located primarily in the nucleus, HDAC9 is broadly expressed, with highest levels in brain, heart, muscle, and testis. Isoform 3 is detected in bladder carcinoma cells.
  • The protein is subject to alternative splicing, phosphorylation, and ubiquitination, and contains a zinc-binding domain essential for its hydrolase activity.
