- HDAC9 is a histone deacetylase that catalyzes the removal of acetyl groups from lysine residues on histones H2A, H2B, H3, and H4, leading to transcriptional repression and epigenetic regulation.
- It represses MEF2-dependent transcription, playing critical roles in cell cycle progression, developmental events, and skeletal myogenesis inhibition.
- Isoform 3 lacks catalytic active site residues and is enzymatically inactive; it represses transcription by recruiting HDAC1 and/or HDAC3.
- HDAC9 protects neurons from apoptosis by inhibiting JUN phosphorylation via MAPK10 and repressing JUN transcription through HDAC1 recruitment.
- Located primarily in the nucleus, HDAC9 is broadly expressed, with highest levels in brain, heart, muscle, and testis. Isoform 3 is detected in bladder carcinoma cells.
- The protein is subject to alternative splicing, phosphorylation, and ubiquitination, and contains a zinc-binding domain essential for its hydrolase activity.

- Because the immunogen is located within the N-terminal region (amino acids 1–100) of human HDAC9, the antibody may recognize multiple isoforms sharing this sequence. Verify isoform specificity in the relevant experimental system.
- For Western blot, a band around 111 kDa is expected for the full-length protein; consider using appropriate loading controls and validate detection in human and rat samples.
- For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), optimize antigen retrieval and antibody concentration, and confirm staining in positive control tissues such as brain or heart.
