Biological Background: HARS2 (Mitochondrial Histidine--tRNA Ligase) Function and Localization
- HARS2 encodes mitochondrial histidine--tRNA ligase, also known as histidyl-tRNA synthetase (HisRS) and histidine--tRNA ligase-like. Alternative names include HARSL, HARSR, HO3, and PRLTS2.
- This enzyme belongs to the class II aminoacyl-tRNA synthetase family and catalyzes the ATP-dependent attachment of histidine to its cognate tRNA(His), forming histidyl-tRNA(His) and AMP.
- HARS2 is localized to the mitochondrion, where it plays a critical role in mitochondrial protein translation by providing charged tRNA(His) for ribosome-mediated polypeptide elongation.
- High expression is observed in heart, kidney, and skeletal muscle, with lower levels in brain and liver, reflecting tissue-specific mitochondrial activity.
- Post-translational modifications include acetylation and phosphorylation, which may regulate enzyme activity or stability (Keywords: Acetylation, Phosphoprotein).
- Mutations in HARS2 are associated with Perrault syndrome 2 (PRLTS2), a disorder involving sensorineural hearing loss and ovarian dysfunction, underscoring its clinical significance in mitochondrial disease.
Experimental Guidance and Technical Tips
- For WB, a band around 57 kDa is expected, but note that endogenous cleavage or post-translational modifications may result in additional bands. Validate in relevant sample systems.
- As a polyclonal antibody, it is well-suited for ELISA due to its ability to bind multiple epitopes; consider optimizing coating and detection conditions.
- The immunogen corresponds to a region conserved among human, mouse, and rat, supporting cross-reactivity. Validate specificity in samples from these species to confirm.
- Since HARS2 is mitochondrial, ensure lysis buffers are compatible with mitochondrial protein extraction if targeting endogenous protein in tissues.
