Biological Background: GSTT1 Function and Localization

• GSTT1 encodes glutathione S-transferase theta-1 (GST class-theta-1), also known as glutathione transferase T1-1. Alternative splicing of the gene has been reported.
• The enzyme catalyzes the conjugation of reduced glutathione to a wide range of exogenous and endogenous hydrophobic electrophiles, including 1,2-epoxy-3-(4-nitrophenoxy)propane, phenethylisothiocyanate, 4-nitrobenzyl chloride, and 4-nitrophenethyl bromide.
• GSTT1 also exhibits glutathione peroxidase activity, reducing cumene hydroperoxide and potentially contributing to cellular protection against oxidative stress.
• Subcellularly, GSTT1 is localized in the cytoplasm, where it participates in phase II detoxification reactions.
• Tissue expression is found in erythrocytes, low levels in liver, and in lung at the alveolar/bronchiolar junction in club cells and ciliated cells, but the protein is absent from epithelial cells of larger bronchioles.
