Biological Background: GSTT1 Function and Localization
- GSTT1 encodes glutathione S-transferase theta-1 (GST class-theta-1), also known as glutathione transferase T1-1. Alternative splicing of the gene has been reported.
- The enzyme catalyzes the conjugation of reduced glutathione to a wide range of exogenous and endogenous hydrophobic electrophiles, including 1,2-epoxy-3-(4-nitrophenoxy)propane, phenethylisothiocyanate, 4-nitrobenzyl chloride, and 4-nitrophenethyl bromide.
- GSTT1 also exhibits glutathione peroxidase activity, reducing cumene hydroperoxide and potentially contributing to cellular protection against oxidative stress.
- Subcellularly, GSTT1 is localized in the cytoplasm, where it participates in phase II detoxification reactions.
- Tissue expression is found in erythrocytes, low levels in liver, and in lung at the alveolar/bronchiolar junction in club cells and ciliated cells, but the protein is absent from epithelial cells of larger bronchioles.
- Post-translational modifications and structural features have been studied, with direct protein sequencing and 3D-structure determination, supporting its classification as a theta-class transferase.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant protein covering amino acids 1-240 of human GSTT1 (NP_000844.2). This full-length immunogen suggests the antibody recognizes an epitope within the native protein, but experimental validation is recommended.
- For Western blot (WB), consider using whole cell lysates from human, mouse, or rat tissues or cell lines known to express GSTT1, such as liver or lung samples. A band around 27 kDa is expected.
- The antibody is also suitable for ELISA; preliminary titration and optimization of antigen coating and detection conditions will help achieve optimal signal-to-noise ratios.
- Since the antibody cross-reacts with mouse and rat GSTT1, it can be used in comparative studies, but specificity should be confirmed for each species and application.
