Biological Background: GRHL2 Function and Localization

  • GRHL2 (Grainyhead-like protein 2 homolog), also known as Brother of mammalian grainyhead (BOM) and Transcription factor CP2-like 3 (TFCP2L3), is a member of the grainyhead-like transcription factor family that includes GRHL1, GRHL2, and GRHL3.
  • Plays a critical role in primary neurulation and epithelial development by acting as both a transcriptional activator and repressor, binding to the consensus DNA sequence 5'-AACCGGTT-3'. Related references: PMID:25152456, PMID:29309642
  • Cooperates with GRHL3 to establish distinct zones of primary neurulation; essential for closure 3 (rostral forebrain) and posterior neuropore closure.
  • Regulates epithelial morphogenesis by up-regulating tight junction components CLDN3 and CLDN4, and enhances CLDN4 localization at tight junctions via RAB25.

  • The immunogen corresponds to amino acids 110–210 of human GRHL2, a region that may be conserved across species; given reported cross-reactivity with human and rat, validation in other species (e.g., mouse) should be performed if required.
  • For Western blotting, use whole-cell lysates from GRHL2-expressing cells such as HaCaT (human keratinocytes), placental tissue, or prostate cancer cell lines. The antibody is expected to detect bands at approximately 69 kDa and 71 kDa, corresponding to the two major isoforms.
  • For ELISA, consider the application’s requirement for native versus denatured protein recognition, and include appropriate controls.
