Biological Background: GRB10 Function and Localization

  • GRB10 (Growth factor receptor-bound protein 10), also known as Grb10 adapter protein or insulin receptor-binding protein Grb-IR, is encoded by the GRB10 gene (human GeneID: 2887) and belongs to the GRB7/10/14 family of adapter proteins.
  • Acts as an adapter protein modulating coupling of activated receptor tyrosine kinases with downstream signaling pathways; specifically binds to and suppresses signals from insulin (INSR) and insulin-like growth factor (IGF1R) receptors via two mechanisms: interference with signaling and promotion of receptor degradation.
  • Negatively regulates insulin signaling by delaying and reducing AKT1 phosphorylation, blocking INSR association with IRS1/IRS2, and preventing insulin-stimulated IRS1/IRS2 tyrosine phosphorylation.
  • Mediates ubiquitination-dependent receptor downregulation by recruiting NEDD4 to IGF1R, leading to increased internalization and degradation via proteasomal and lysosomal pathways; may also promote INSR ubiquitination and proteasomal degradation.
  • Negatively regulates Wnt signaling by interacting with the intracellular portion of LRP6 and interfering with AXIN1 binding to LRP6.
  • Positively regulates the KDR/VEGFR-2 signaling pathway, possibly by inhibiting NEDD4-mediated degradation of KDR.
  • Expressed widely in fetal and adult tissues, including liver, lung, kidney, skeletal muscle, heart, spleen, skin, and brain; subcellular localization is primarily cytoplasmic.
  • Features a C-terminal SH2 domain and is subject to phosphorylation and alternative splicing (UniProt Keywords: Alternative splicing, Phosphoprotein, SH2 domain).

  • The immunogen is a synthetic peptide from the N-terminal region (aa 1-100) of human GRB10. For WB, consider using a reduced denaturing gel and validate with positive controls from human cell lines known to express GRB10 (e.g., HepG2, HEK293).
