Biological Background: Granulin Function and Localization
- Progranulin (PGRN), also known as acrogranin, epithelin precursor, GP88, and PCDGF, is a secreted glycoprotein that acts as a key regulator of lysosomal function and as a growth factor involved in inflammation, wound healing, and cell proliferation.
Related references: PMID:12526812 PMID:18378771 PMID:28073925 - PGRN regulates protein trafficking to lysosomes and lysosomal enzyme activity, and facilitates lysosomal acidification leading to CTSD degradation.
Related references: PMID:28453791 PMID:28541286 PMID:28073925 - As a wound-related growth factor, it promotes dermal fibroblast and endothelial cell division, migration, and capillary-like tube formation, and enhances epithelial cell proliferation by blocking TNF-mediated neutrophil activation.
Related references: PMID:12526812 - In neurons, progranulin modulates inflammation and preserves neuronal survival, axonal outgrowth, and integrity.
Related references: PMID:18378771 - Subcellularly, progranulin is secreted and also localizes to lysosomes.
- It is highly expressed in kidney and present in inflammatory cells, bone marrow, and epithelial cell lines; additional functions include promoting A431 cell proliferation, inhibiting epithelial cell proliferation and inducing IL-8 secretion, and stabilizing CTSD via direct interaction.
Experimental Guidance and Technical Tips
- The immunogen maps to amino acids 494–593 in the C-terminal region of human progranulin; verify sequence alignment with mouse to confirm expected cross-reactivity.
- For western blotting, full-length progranulin migrates near 64 kDa, but proteolytic processing yields smaller granulin peptides; include appropriate controls and validate in the relevant sample system.
- In IF/ICC, permeabilization steps may be necessary to visualize lysosomal pools; consider co-staining with organelle markers.
- As a secreted protein, progranulin may be enriched in serum-free conditioned medium or after stimulation; optimize sample preparation for ELISA assays.
