  • Platelet glycoprotein VI (GP6) is a type I transmembrane glycoprotein and a member of the immunoglobulin superfamily.
  • It functions as the major collagen receptor on platelets, mediating collagen-induced platelet adhesion, activation, and aggregation.
  • Upon ligand binding, GP6 signals through the immunoreceptor tyrosine-based activation motif (ITAM)-coupled FcRγ chain, leading to activation of Src kinases, SYK, LAT, and PLCγ2.
  • GP6 plays a pivotal role in platelet procoagulant activity, promoting thrombin generation and fibrin formation, thereby contributing to both arterial and venous thrombosis.
  • The protein is predominantly expressed on the plasma membrane of megakaryocytes and platelets.
  • Alternative splicing generates multiple GP6 isoforms.
  • Post-translational modifications include glycosylation and disulfide bond formation.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal extracellular domain (aa 21–267), which includes the ligand-binding immunoglobulin-like domains. This region is well-suited for detecting both full-length and soluble GP6 fragments.
  • For Western blot, the predicted molecular weight of human GP6 is ~37 kDa; however, glycosylation may cause a shift in apparent molecular weight. Validate in platelet lysates or GP6-expressing cell lines.
  • The antibody cross-reacts with mouse and rat GP6, enabling studies in rodent models of thrombosis and hemostasis.
  • In ELISA, consider using recombinant GP6 extracellular domain as a positive control to assess binding.
  • For immunohistochemistry or flow cytometry, note that this polyclonal antibody may recognize both native and denatured GP6; optimization is recommended.
