Biological Background: GORASP2 Function and Localization
- GORASP2 (also known as GRASP55, GOLPH6, p59, GRS2) is a key structural protein of the Golgi apparatus that interacts with GORASP1/GRASP65 to form and maintain the Golgi ribbon. Related references: PMID:33301566
- It plays a role in the assembly and membrane stacking of Golgi cisternae, particularly during the reassembly of Golgi stacks after mitosis and meiosis. Related references: PMID:10487747 PMID:21515684 PMID:22523075
- Mediates ER stress-induced unconventional trafficking of core-glycosylated CFTR to the plasma membrane, bypassing the Golgi. Related references: PMID:21884936 PMID:27062250 PMID:28067262
- Regulates the intracellular transport and cell surface presentation of transmembrane proteins such as TGFA. Related references: PMID:11101516
- Required for normal acrosome formation during spermiogenesis and male fertility, likely by facilitating JAM2 and JAM3 localization at germ cell–Sertoli cell contact sites.
- GORASP2 is localized to the Golgi apparatus membrane and endoplasmic reticulum membrane, and is subject to phosphorylation, myristoylation, palmitoylation, and methylation.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a C-terminal region (aa 353-452) of human GORASP2; based on cross-reactivity data, this region is conserved in mouse and rat.
- For western blotting, reduce and denature samples; expect a band around 47 kDa. Use appropriate positive controls (e.g., HeLa or HEK293 cell lysates) and validate specificity in your system.
- For immunofluorescence/immunocytochemistry, fix cells with paraformaldehyde and permeabilize; anticipate Golgi-like perinuclear staining. Co-staining with a Golgi marker (e.g., GM130) is recommended to confirm localization.
- As a polyclonal antibody, lot-to-lot consistency should be verified for critical assays; titrate the antibody for each new application.
