Biological Background: GNAI2 Function and Localization
- GNAI2 encodes the guanine nucleotide-binding protein G(i) subunit alpha-2, which acts as a transducer in GPCR signaling cascades, coupling receptor activation to intracellular effectors [PMID:38505600, 29925945, 35637350].
Related references: PMID:38505600, PMID:29925945, PMID:35637350 - The alpha subunit alternates between an inactive GDP-bound state and an active GTP-bound state; upon GPCR activation, GDP is released and replaced by GTP, triggering downstream signaling [PMID:12359238, 29925945].
Related references: PMID:12359238, PMID:29925945 - GNAI2 inhibits adenylate cyclase activity, reducing cellular cAMP levels, and mediates inhibitory regulation of hormone-sensitive adenylate cyclase in response to beta-adrenergic stimuli.
- In endothelial cells, GNAI2 plays an important role in activating the transcription factor NFAT to promote angiogenesis [PMID:38505600].
Related references: PMID:38505600 - GNAI2 is essential for neutrophil recruitment during acute inflammation, facilitating polymorphonuclear neutrophil transmigration (By similarity).
- It regulates the cell surface density of dopamine receptors DRD2 by sequestering them as an intracellular pool.
- GNAI2 localizes to the cytoplasm, centrosome, and cell membrane, reflecting its roles in signal transduction and cytoskeletal interactions.
- The protein is highly expressed in endothelial cells and undergoes lipid modifications (myristoylation and palmitoylation) that anchor it to membranes.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 250–355 of human GNAI2; therefore, the antibody is likely to detect a linear epitope and is well-suited for WB under denaturing and reducing conditions.
- For IHC-P, consider using heat-induced antigen retrieval in citrate buffer (pH 6.0) to expose the epitope in formalin-fixed paraffin-embedded tissues.
- Validate antibody specificity using appropriate positive (e.g., human endothelial cells or tissues with high GNAI2 expression) and negative controls; note the predicted molecular weight of ~40 kDa on Western blots.
- Optimize antibody dilutions for each application and sample type; a starting range of 1:500–1:2000 for WB and 1:50–1:200 for IHC-P is commonly used, but empirical determination is recommended.
