Biological Background: GM130/GOLGA2 Function and Localization
- GM130, encoded by the GOLGA2 gene, is a 130 kDa cis-Golgi matrix protein also known as golgin-95 and GM130 autoantigen (UniProt Q08379).
- Acts as a peripheral membrane component of the cis-Golgi, maintaining Golgi stack structure and facilitating vesicle tethering and fusion to the Golgi membrane (PubMed:16489344).
Related references:
PMID:16489344 - Together with p115/USO1 and STX5, it mediates vesicle docking and fusion to maintain an interconnected, stacked Golgi apparatus (By similarity).
- Plays a critical role in mitotic Golgi disassembly: phosphorylation at Ser-37 by CDK1 during mitosis disrupts interaction with p115, inhibiting COPI vesicle tethering and Golgi transport (By similarity).
- Also essential for spindle pole assembly and centrosome organization; upon mitotic phosphorylation, it recruits importin-α to Golgi membranes, releasing TPX2 to activate AURKA and nucleate microtubules, thereby linking mitotic membranes to the spindle (PubMed:19242490, PubMed:26165940).
Related references:
PMID:19242490, PMID:26165940 - Required for Golgi ribbon formation and efficient glycosylation of membrane and secretory proteins (PubMed:16489344, PubMed:17314401).
Related references:
PMID:16489344, PMID:17314401 - Localizes primarily to the cis-Golgi network membrane and the endoplasmic reticulum-Golgi intermediate compartment, and under mitotic conditions also associates with the spindle pole and cytoskeleton.
- Annotated with keywords including Alternative splicing, Cell cycle, Cell division, Coiled coil, Phosphoprotein, and Microtubule, reflecting its diverse regulatory roles.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 1–100 of human GM130; the antibody may preferentially detect targets containing this N-terminal region.
- For Western blotting, a band at approximately 113–130 kDa is expected; include a known positive control (e.g., HeLa or HEK293 cell lysate) and adjust antibody dilution accordingly.
- In immunofluorescence/ICC, anticipate perinuclear cis-Golgi staining; co-staining with a cis-Golgi marker (e.g., GM130 itself as a known marker) can aid interpretation.
- For immunohistochemistry (IHC-P), test antigen retrieval methods and optimize primary antibody concentration using tissues with known GM130 expression.
- The antibody is also suitable for ELISA; users may consider sandwich ELISA designs using paired antibodies if available.
