- Glycogen synthase 1 (GYS1), also known as muscle glycogen synthase, is the key enzyme in glycogen biosynthesis, catalyzing the transfer of glucosyl residues from UDP-glucose to the non-reducing end of alpha-1,4-glucan primers.
- It works in concert with glycogenin (which initiates the glycogen primer) and glycogen branching enzyme to extend and branch the glycogen molecule.
- GYS1 is expressed predominantly in skeletal muscle, but also in most cell types where glycogen serves as an energy reservoir.

- For Western blotting, use reducing SDS-PAGE and load 20–50 µg of total protein from skeletal muscle, liver, or cell line lysates known to express GYS1. The predicted band size is approximately 84 kDa.
- When performing ELISA, consider using a paired antibody format if developing a sandwich assay, and validate the antibody’s performance with recombinant GYS1 protein or cell lysates to confirm sensitivity and specificity.
- Since GYS1 is a phosphoprotein, treatment of samples with phosphatase inhibitors may be necessary to preserve phosphorylation states if those are targets of interest.
