Biological Background: GLI1 Function and Localization
-
GLI1 is a zinc-finger transcription activator and key effector of the hedgehog signaling pathway, essential for craniofacial, digital, CNS, and gastrointestinal development. It binds the DNA consensus sequence 5'-GACCACCCA-3' to activate target genes. Related references: PMID:10806483, PMID:11238441, PMID:19706761, PMID:2105456, PMID:24217340, PMID:2832761
-
Unlike related family members GLI2 and GLI3, GLI1 lacks a repressor domain and acts exclusively as a transcriptional activator. Related references: PMID:10075717
-
An alternative splice isoform (isoform 2) activates a distinct gene set, including CD24, and displays restricted tissue expression compared to the canonical isoform 1. Related references: PMID:19706761
-
GLI1 shuttles between the cytoplasm and nucleus, and localizes to primary cilia. Hedgehog pathway activation promotes its nuclear accumulation.
-
GLI1 protein is detected in testis, myometrium, fallopian tube, and specific brain regions (cerebellum, optic nerve, olfactory tract). In contrast to normal adult tissues, where it is largely undetectable, GLI1 is highly expressed in various cancers. Related references: PMID:2105456, PMID:19878745
Experimental Guidance and Technical Tips
- For Western blot, GLI1 migrates at approximately 118 kDa. Ensure complete transfer of high molecular weight proteins and include appropriate loading controls.
- In IHC-P and IF/ICC, expect nuclear and cytoplasmic staining; signal intensity may correlate with hedgehog pathway activity in the tissue.
- This antibody cross-reacts with mouse and rat GLI1, supporting preclinical studies; however, validate under your specific fixation and antigen retrieval protocols.
- The immunogen corresponds to the N-terminal region (aa 1–233); confirm isoform detection and specificity in your assay system if studying particular splice variants.
- For ELISA, pair with a suitable secondary detection antibody and empirically determine optimal working concentrations.
