Biological Background: GGT1 Function and Localization
- GGT1 is synthesized as a 61 kDa proenzyme (UniProt: 61410 Da) that is processed into a 39 kDa heavy chain and a 24 kDa light chain, which associate to form the active enzyme.
- It catalyzes the cleavage of the gamma-glutamyl bond of extracellular glutathione (GSH) and glutathione conjugates, releasing glutamate and cysteinyl-glycine, thereby regulating glutathione homeostasis and cysteine supply. Related references: PMID:21447318, PMID:27791009
- GGT1 converts leukotriene C4 (LTC4) to LTD4, a critical step in the synthesis of cysteinyl leukotrienes, which are potent pro-inflammatory mediators. Related references: PMID:17924658, PMID:21447318, PMID:7673200
- The enzyme can also catalyze transpeptidation reactions, transferring the gamma-glutamyl group to dipeptides or amino acids to form novel gamma-glutamyl compounds. Related references: PMID:17924658, PMID:21447318, PMID:7673200
- GGT1 is a type II transmembrane protein anchored to the plasma membrane via an N-terminal signal-anchor domain, with its catalytic domain exposed to the extracellular space.
- The protein is heavily glycosylated and contains sialic acid modifications, which are critical for its proper folding, stability, and enzymatic function.
- It is highly expressed in kidney and liver, as well as in pancreas, stomach, intestine, placenta, and lung; alternative promoter usage yields tissue-specific isoforms.
- A lung-specific isoform appears to be catalytically inactive, suggesting tissue-dependent functional regulation.
Experimental Guidance and Technical Tips
- The immunogen was generated against a recombinant fusion protein corresponding to amino acids 370–569 of human GGT1 (NP_038265.2), a region located in the C-terminal light chain. This design suggests the antibody may detect the 24 kDa light chain, the 61 kDa proenzyme, and possibly the 39 kDa heavy chain depending on sample processing and gel conditions.
- For western blotting, positive control samples could include human kidney or liver lysates, which naturally express high levels of GGT1.
- In IHC-P applications, anticipate plasma membrane staining; antigen retrieval conditions may need optimization for your tissue type and fixation method.
- This antibody has reported cross-reactivity with mouse and rat GGT1, but users should validate species-specific reactivity and assay conditions independently.
- Due to the multi-subunit nature of GGT1, consider running reduced and non-reduced samples to characterize band patterns.
