Biological Background: GCN2 Function and Localization
- GCN2 (eIF-2-alpha kinase 4, also known as GCN2-like protein) is a member of the eIF-2-alpha kinase family, functioning as a key metabolic-stress sensor.
- Phosphorylates the alpha subunit of eukaryotic translation initiation factor 2 (eIF-2-alpha) in response to amino acid starvation, leading to global attenuation of cap-dependent translation while promoting translation of stress-responsive mRNAs such as ATF4. - Acts as an activator of the integrated stress response (ISR), essential for cellular adaptation to amino acid deprivation.
