  • GCET2, also known as HGAL (human germinal center-associated lymphoma) and GCAT2, is encoded by the GCSAM gene. It is specifically expressed in germinal center B-cells and lymphomas, playing a critical role in B-cell biology.
  • The protein functions as a negative regulator of lymphocyte motility. It mediates the migration-inhibitory effects of interleukin-6 (IL-6), thereby controlling lymphocyte trafficking within lymphoid organs.
  • GCET2 positively regulates the RhoA signaling pathway, enhancing RhoA activation and subsequent ROCK kinase activity. This leads to inhibition of lymphocyte and lymphoma cell motility.
  • As a regulator of B-cell receptor signaling, GCET2 acts through SYK kinase activation, influencing downstream signaling events in B-cells.
  • Subcellularly, GCET2 localizes to both the cytoplasm and cell membrane, indicating dynamic distribution that may be critical for its signaling functions.
  • The protein undergoes alternative splicing and is post-translationally modified by phosphorylation and ubiquitin conjugation (Ubl), as indicated by Phosphoprotein and Ubl conjugation keywords.
  • Tissue expression is prominent in normal germinal center lymphocytes, thymus, spleen, and germinal center-derived malignancies, including diffuse large B-cell lymphoma (DLBCL).

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (aa 1–178) of human GCET2, which is conserved across species; thus, the antibody may detect human, mouse, and rat GCET2 in Western blot and ELISA. Validate cross-reactivity in your assay system.
  • For Western blot, a band at approximately 21 kDa is expected. Due to potential post-translational modifications or isoform variability, additional bands might appear; verify with appropriate controls.
  • In ELISA, consider using recombinant GCET2 protein or cell lysates from germinal center B-cell lines as antigen; optimize coating and detection conditions experimentally.
  • The antibody is produced in rabbit as a monoclonal reagent, offering batch-to-batch consistency for quantitative assays.
