Biological Background: Galectin-3 Function and Localization

  • Galectin-3 (also known as 35 kDa lectin, CBP35, GALBP, IgE-binding protein, L-31, Laminin-binding protein, Lectin L-29, and Mac-2 antigen) is a galactose-specific lectin encoded by the LGALS3 gene in humans.
  • It binds IgE and mediates CSPG4-stimulated endothelial cell migration via interaction with alpha-3, beta-1 integrin. Together with DMBT1, it is required for terminal differentiation of columnar epithelial cells during early embryogenesis (By similarity).
  • In the nucleus, Galectin-3 functions as a pre-mRNA splicing factor, participating in mRNA processing and splicing.
  • It plays a role in acute inflammatory responses, including neutrophil activation and adhesion, chemoattraction of monocytes/macrophages, opsonization of apoptotic neutrophils, and mast cell activation.
  • Together with TRIM16, Galectin-3 coordinates the recognition of damaged endomembranes with mobilization of core autophagy regulators ATG16L1 and BECN1, linking innate immunity to autophagy.
  • The protein localizes to the cytoplasm, nucleus, and is also secreted. Major expression is found in colonic epithelium, activated macrophages, and fetal membranes.
  • Keywords associated with Galectin-3 include acetylation, phosphorylation, disulfide bond, immunity, lectin, spliceosome, and proteomics identification.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide corresponding to amino acids 1–100 of human Galectin-3. This N-terminal region is distinct from the carbohydrate-recognition domain and may aid in detecting both full-length and cleaved forms.
  • For Western blotting, the predicted molecular weight is approximately 26 kDa, but due to post-translational modifications and the protein’s propensity to form multimers, both monomeric and higher molecular weight bands may be observed; consider including reducing agents in sample buffers.
  • In immunohistochemistry (IHC-P), note that Galectin-3 can be cytoplasmic, nuclear, or secreted; antigen retrieval methods may be required, and appropriate controls should be included to confirm staining specificity.
  • The antibody cross-reacts with mouse and rat Galectin-3, making it suitable for translational studies across these species; however, validate expression levels in the target tissue before quantitative comparisons.
  • For immunoprecipitation (IP) and ELISA, the monoclonal nature of the antibody provides consistent batch-to-batch performance; optimize antibody concentration empirically for each assay format.
  • Always include appropriate positive and negative controls, such as known Galectin-3-expressing cell lines (e.g., macrophages, colonic epithelial cells) and knockout or knockdown models where possible.
