Biological Background: GABA transaminase (ABAT) Function and Localization
-
ABAT encodes the mitochondrial enzyme 4-aminobutyrate aminotransferase, also known as GABA aminotransferase (GABA-AT), (S)-3-amino-2-methylpropionate transaminase, and L-AIBAT (UniProt: P80404).
-
Catalyzes the conversion of gamma-aminobutyrate (GABA) to succinate semialdehyde, a key step in neurotransmitter degradation; also acts on L-beta-aminoisobutyrate to produce methylmalonate semialdehyde. Related references: PMID:10407778 PMID:15528998
-
Can utilize delta-aminovalerate and beta-alanine as substrates (By similarity).
-
Localizes to the mitochondrial matrix, imported via an N-terminal transit peptide.
-
Contains a pyridoxal phosphate cofactor and an iron-sulfur cluster essential for catalytic activity (UniProt keywords: Aminotransferase, Iron, Iron-sulfur, Pyridoxal phosphate, Transferase).
-
Post-translational modifications include acetylation and disulfide bond formation (UniProt keywords: Acetylation, Disulfide bond).
-
Tissue expression: highest in liver, followed by pancreas, brain, kidney, heart, and placenta.
-
Listed as a disease variant; mutations in ABAT are linked to neurological disorders (UniProt keyword: Disease variant).
Experimental Guidance and Technical Tips
-
The immunogen spans the N-terminal region (amino acids 1-100), which includes the mitochondrial transit peptide; the antibody may detect both precursor and mature forms. Consider this banding pattern in WB.
-
For Western blotting, the predicted molecular weight is ~56 kDa. Use an appropriate loading control and validate specificity with positive/negative lysates.
-
In IHC-P, antigen retrieval may be necessary; include isotype controls and verify staining patterns relative to known tissue expression profiles.
-
For ELISA, the antibody can serve as a capture or detection reagent; titrate under assay-specific conditions and confirm sensitivity with recombinant antigen.
-
Cross-reactivity with mouse and rat is indicated; when using non-human samples, validate performance under your experimental conditions.
