Biological Background: Tyrosine-protein kinase Fyn Function and Localization

  • Tyrosine-protein kinase Fyn is a member of the Src family of non-receptor tyrosine kinases, characterized by SH2 and SH3 protein interaction domains and an N-terminal myristoylation/palmitoylation site for membrane localization.
  • Plays a broad role in signaling pathways regulating cell growth, survival, adhesion, motility, and cytoskeletal organization, acting downstream of various receptors. Related references: PMID:11536198 PMID:15489916 PMID:15557120
  • Inactive FYN is phosphorylated on its C-terminal tail; upon activation, it associates with and phosphorylates PTK2/FAK1, promoting focal adhesion turnover and cell migration. Related references: PMID:15489916
  • Regulates cytoskeletal dynamics by phosphorylating substrates such as WAS (actin regulator), MAP2, and MAPT (microtubule-associated proteins). Related references: PMID:14707117 PMID:15536091
  • In the immune system, Fyn participates in T-cell receptor (TCR) signaling, promoting T-cell differentiation and proliferation. It phosphorylates PTK2B/PYK2 upon TCR activation and mediates CD28-induced phosphorylation of VAV1. Related references: PMID:22080863 PMID:20028775 PMID:11005864
  • Provides negative feedback in TCR signaling by phosphorylating PAG1, which recruits CSK to lipid rafts to inactivate SFKs, and PDCD1, leading to SHP-2 recruitment and dephosphorylation of proximal signaling molecules. Related references: PMID:18056706 PMID:32184441
  • In the nervous system, Fyn is highly expressed and phosphorylates proteins critical for axon guidance and synaptic function, including DPYSL2, ARHGAP32, and SNCA (alpha-synuclein). Related references: PMID:11162638 PMID:12788081 PMID:19652227
  • Isoform 1 is the predominant form in the brain, while isoform 2 is expressed in hematopoietic cells, particularly T-lymphocytes. Subcellular localization includes the cytoplasm, nucleus, cell membrane, and perikaryon.

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide corresponding to amino acids 200–300 of human Fyn (NP_002028.1); the antibody may recognize epitopes within this region. Validate specificity in your experimental system.
  • For western blotting (WB), detect Fyn at a predicted molecular weight of 61 kDa. Consider loading appropriate positive controls from human, mouse, or rat lysates.
  • In immunohistochemistry, test the antibody on formalin-fixed paraffin-embedded (IHC-P) or frozen sections (IF-P, IF/ICC) with appropriate antigen retrieval and blocking protocols.
  • The antibody cross-reacts with mouse and rat Fyn, making it suitable for studies in these species.
  • For ELISA, use as per standard indirect assay protocols; consider pairing with a detection antibody for quantitative applications.
