Biological Background: FUT3 Function and Localization

FUT3 (Fucosyltransferase 3, also known as FucT-III, CD174, Lewis FT) is a Golgi membrane-bound glycosyltransferase responsible for the synthesis of Lewis blood group antigens, including Lewis a, Lewis b, Lewis x (SSEA-1), and Lewis y, through the transfer of L-fucose to type 1 and type 2 oligosaccharide acceptors. It catalyzes alpha(1,4)-fucosylation of type 1 chains (Galβ1-3GlcNAc) and alpha(1,3)-fucosylation of type 2 chains (Galβ1-4GlcNAc), with a preference for type 1 acceptors, and can also utilize sialylated precursors to generate sialyl Lewis a (sLe^a) and sialyl Lewis x (sLe^x) epitopes. Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the N-terminal region (aa 1–100) of human FUT3; therefore, the antibody may recognize both native and denatured forms. For Western blot, a band at approximately 42 kDa is expected. Consider using standard reducing conditions and a suitable blocking agent.
- For ELISA, validate the antibody with the immunogen peptide or recombinant FUT3 protein to confirm reactivity. Pair with an appropriate secondary antibody for detection.
- Cross-reactivity with rat is reported. Validate in the relevant sample system and consider sequence alignment to assess potential recognition of other FUT family members.
