Biological Background: FOXP1 Function and Localization
- FOXP1 is a transcriptional repressor that can synergize with CTBP1 to repress transcription, though CTBP1 is not essential. Related references: PMID:18347093 PMID:26647308
- It plays a critical role in lung epithelium specification and differentiation, cooperating with FOXP4 to regulate secretory cell fate by restricting goblet cell lineage.
- Acts as an essential transcriptional regulator of B-cell development and can repress pro-apoptotic genes to promote B-cell expansion via NF-kappa-B signaling. Related references: PMID:25267198
- Negatively regulates the differentiation of T follicular helper cells and is involved in monocyte/macrophage differentiation by binding to the CSF1R promoter. Related references: PMID:15286807 PMID:18347093 PMID:18799727
- Involved in spinal motor neuron organization, promoting formation of lateral motor neuron column and preganglionic motor column.
- Participates in cardiac muscle cell proliferation regulation and angiogenesis, where it suppresses SEMA5B to promote endothelial cell functions. Related references: PMID:24023716
- Isoform 8 is specifically expressed in embryonic stem cells and stimulates pluripotency gene expression while repressing differentiation genes. Related references: PMID:21924763
- Localized to the nucleus, FOXP1 contains zinc-finger domains and undergoes post-translational modifications including phosphorylation and ubiquitination.
Experimental Guidance and Technical Tips
- The immunogen (aa 550-677) includes part of the forkhead DNA-binding domain, which is highly conserved across human, mouse, and rat, supporting the observed cross-reactivity.
- For Western blotting, expect a band around 75 kDa; note that isoform 8 (embryonic stem cell-specific) may show a different size.
- Validate antibody specificity in your experimental system, as FOXP1 has multiple isoforms and tissue-specific expression.
- For immunohistochemistry (paraffin), optimize antigen retrieval; nuclear staining is anticipated.
- For ELISA, perform titration and include appropriate positive and negative controls.
