Biological Background: FOXL2 Function and Localization

  • FOXL2 is a forkhead box transcription factor critical for ovary differentiation and maintenance, and functions as a repressor of the somatic testis determination program.
  • It prevents ovary-to-testis trans-differentiation through transcriptional repression of the Sertoli cell-promoting gene SOX9 (by similarity).
  • FOXL2 has apoptotic activity in ovarian cells and modulates estrogen signaling by suppressing ESR1-mediated PTGS2/COX2 transcription and regulating CYP19 expression (by similarity).
  • It participates in SMAD3-dependent transcription of FST and activates SIRT1 under cellular stress conditions, as well as OSR2 transcription.
  • FOXL2 is localized exclusively in the nucleus.
  • It is expressed early in the somatic cells of the developing gonad and persists in follicular cells of the adult ovary, in addition to developing eyelid.
  • The protein is a phosphoprotein and is modified by ubiquitin conjugation (isopeptide bond); it is associated with premature ovarian failure and features a triplet repeat expansion.
