Biological Background: FNDC5 Function and Localization
- FNDC5, also known as fibronectin type III domain-containing protein 5 or irisin, is a membrane protein that can be proteolytically cleaved to release the irisin hormone.
- Irisin mediates beneficial effects of muscular exercise, notably inducing the browning of white adipose tissue by stimulating UCP1 expression, at least partly via the nuclear receptor PPARA.
- The protein belongs to the fibronectin type III domain-containing family and is encoded by the FNDC5 gene (also known as FRCP2) on chromosome 1.
- Subcellularly, full-length FNDC5 localizes to the cell membrane and peroxisome membrane, while the cleaved irisin portion is secreted.
- FNDC5 is widely expressed, with highest levels detected in the heart; expression is very low or absent in colon, pancreas, and spleen.
- Post-translational modifications include glycosylation, and the protein is subject to alternative promoter usage and alternative splicing, generating multiple isoforms.
- As a hormone and transmembrane protein, FNDC5 plays a critical role in energy metabolism and exercise physiology.

- The immunogen is a synthetic peptide spanning amino acids 1–150 (N-terminal region) of human FNDC5. For WB, consider testing on heart or skeletal muscle lysates, where expression is high.
- In IF/ICC, detection may require permeabilization for membrane-bound FNDC5, while irisin secretion can be monitored in culture supernatants via ELISA.
- Cross-reactivity is predicted for human, mouse, and rat; validate in the relevant sample system before routine use.
- The predicted molecular weight of full-length FNDC5 is ~24 kDa, but cleaved irisin (~12 kDa) may also be observed; use appropriate controls.
