Biological Background: FES Function and Localization
- FES encodes the tyrosine-protein kinase Fes/Fps, a non-receptor protein tyrosine kinase belonging to the Fes/Fer family. Also known as feline sarcoma/Fujinami avian sarcoma oncogene homolog, proto-oncogene c-Fes, and p93c-fes.
- It acts downstream of cell surface receptors such as FCER1 and KIT to regulate actin cytoskeleton dynamics, microtubule assembly, cell attachment, and cell spreading.
- Plays a critical role in mast cell signaling, including FCER1-mediated degranulation, and participates in HGF-induced cell scattering and migration through phosphorylation of EZR.
- Functions in cell differentiation and neurite outgrowth in response to NGF signaling, and phosphorylates downstream substrates including BCR, HCLS1/HS1, PECAM1, STAT3, and TRIM28.
- Subcellular localization includes cytoplasm, cytosol, cytoskeleton, cell membrane, cytoplasmic vesicles, Golgi apparatus, and focal adhesions, indicating dynamic spatial regulation.
