Biological Background: FCRL3 Function and Localization

  • FCRL3 (Fc receptor-like protein 3), also known as FcRH3, IRTA3, MAIA, and CD307c, is a type I transmembrane glycoprotein with immunomodulatory roles in both adaptive immunity and fertilization.
  • Promotes TLR9-induced B-cell proliferation, activation, and survival while inhibiting antibody production and suppressing plasma cell differentiation; enhances NF-kB and MAPK signaling pathways. Related references: PMID:23857366

Experimental Guidance and Technical Tips

  • The immunogen (aa 600–720 of human FCRL3) lies within the cytoplasmic tail, so the antibody primarily detects intracellular epitopes; for immunofluorescence, permeabilization may be necessary, but confirm protocol compatibility.
  • For Western blotting, a band at approximately 81 kDa is expected; however, glycosylation and isoform variants may cause slight shifts. Validate with positive controls such as B-cell lines (e.g., Raji, Daudi) or lymphoid tissue lysates.
  • In ELISA, consider using recombinant FCRL3 protein or cell-based assays; optimize coating concentration and detection range empirically.
  • Cross-reactivity with mouse and rat is reported, but due to sequence divergence in the target region, verify specificity and signal intensity in those species.
