Biological Background: FasLG Function and Localization

  • FasLG (CD95L, TNFSF6) is a type II transmembrane protein belonging to the TNF ligand superfamily.
  • It is the canonical ligand for the death receptor FAS (CD95), and this interaction triggers the extrinsic apoptotic pathway crucial for immune cell-mediated cytotoxicity (PubMed:26334989, PubMed:7528780, PubMed:9228058, PubMed:9427603).   • FasLG also binds to the decoy receptor TNFRSF6B/DcR3, which can inhibit FAS-dependent apoptosis (PubMed:27806260). Related references: PMID:27806260
  • Plays an essential role in activation-induced cell death (AICD) of T-cells, contributing to the termination of immune responses and peripheral tolerance (By similarity).
  • Ligation of FAS can additionally activate NF-kappa-B, leading to non-apoptotic signals involved in cell survival and inflammatory responses (By similarity).
