- F4/80, encoded by the Adgre1 (Emr1) gene, is an adhesion G protein-coupled receptor (GPCR) belonging to the EGF-TM7 subfamily.
- The full-length protein is a cell surface glycoprotein with a molecular weight of approximately 102 kDa, containing multiple EGF-like domains and a seven-transmembrane domain.
- F4/80 functions as an orphan receptor thought to mediate cell adhesion and cell-cell interactions, particularly within the immune system.
- It is specifically expressed on the surface of macrophages, with highest levels on mature tissue-resident macrophages, but is absent from macrophages located in T-cell areas of lymph nodes and spleen.
- Low-level expression is also detected on blood monocytes, making F4/80 a well-established pan-macrophage marker in mice.
- The receptor may contribute to the development and function of regulatory T cells (Tregs), linking innate and adaptive immunity.
- Post-translational modifications include glycosylation and disulfide bond formation, which are typical for EGF-like domain-containing proteins and important for structural stability.

- The immunogen consists of a recombinant fragment (aa 367–645) from the extracellular region of mouse F4/80, targeting a sequence with potentially less glycosylation interference; verify that this epitope is exposed in your fixed samples.
- For immunofluorescence (IF) on paraffin-embedded sections (IF-P), consider antigen retrieval with citrate buffer (pH 6.0) or EDTA (pH 9.0) and optimize primary antibody incubation conditions based on the tissue type.
- In immunohistochemistry on paraffin sections (IHC-P), validate the antibody in a known F4/80-positive control (e.g., mouse liver Kupffer cells or splenic red pulp macrophages) and include an isotype control to assess background staining.
- When performing ELISA, use the antibody as a capture or detection reagent against mouse F4/80 recombinant protein and cross-verify reactivity with rat F4/80 if the assay is intended for rat samples.
