Biological Background: EXOSC4 Function and Localization

  • EXOSC4 (also known as RRP41, SKI6, or p12A) is a non-catalytic component of the RNA exosome complex, a multi-subunit machinery with 3′→5′ exoribonuclease activity.
  • In the nucleus, the exosome contributes to the maturation of stable RNA species (rRNA, snRNA, snoRNA) and eliminates processing by-products, pervasive transcripts (PROMPTs), and defective mRNAs.
  • The exosome may participate in immunoglobulin class switch recombination and somatic hypermutation by targeting AICDA deaminase activity to transcribed DNA substrates.
  • In the cytoplasm, the exosome promotes general mRNA turnover, including the degradation of ARE-containing mRNAs and aberrant transcripts in RNA surveillance.
  • EXOSC4 specifically binds to AU-rich elements (AREs) in the 3′ untranslated regions of mRNAs, targeting them for decay.
  • Subcellular localization includes the cytoplasm, nucleolus, nucleoplasm, and nucleus, consistent with its roles in nuclear and cytoplasmic RNA metabolism.
