Biological Background: EXOSC4 Function and Localization
- EXOSC4 (also known as RRP41, SKI6, or p12A) is a non-catalytic component of the RNA exosome complex, a multi-subunit machinery with 3′→5′ exoribonuclease activity.
- In the nucleus, the exosome contributes to the maturation of stable RNA species (rRNA, snRNA, snoRNA) and eliminates processing by-products, pervasive transcripts (PROMPTs), and defective mRNAs.
- The exosome may participate in immunoglobulin class switch recombination and somatic hypermutation by targeting AICDA deaminase activity to transcribed DNA substrates.
- In the cytoplasm, the exosome promotes general mRNA turnover, including the degradation of ARE-containing mRNAs and aberrant transcripts in RNA surveillance.
- EXOSC4 specifically binds to AU-rich elements (AREs) in the 3′ untranslated regions of mRNAs, targeting them for decay.
- Subcellular localization includes the cytoplasm, nucleolus, nucleoplasm, and nucleus, consistent with its roles in nuclear and cytoplasmic RNA metabolism.
- Post-translational modifications of EXOSC4 have been identified, including acetylation, and the protein has been validated by direct protein sequencing and proteomics.

- The immunogen covers amino acids 1–245, which constitutes the full-length human EXOSC4 protein; therefore, the polyclonal antibody may recognize both native and denatured conformations in Western blot.
- For ELISA applications, consider direct antigen coating or capture formats, and optimize antibody concentrations using a titration series.
- Cross-reactivity with rat allows use in rat-derived samples; validation in the specific tissue or cell line of interest is recommended.
- Endogenous EXOSC4 migrates at ~26 kDa; loading controls and molecular weight markers near this size should be included in WB experiments.
