Biological Background: ETF1 Function and Localization

  • ETF1 (Eukaryotic peptide chain release factor subunit 1), also known as ERF1, RF1, or SUP45L1, is a key factor in protein synthesis.
  • Functions within the eRF1-eRF3-GTP ternary complex to recognize stop codons and catalyze peptidyl-tRNA hydrolysis, leading to polypeptide chain release (PubMed:10676813, PubMed:16777602, PubMed:24486019, PubMed:26245381, PubMed:27863242, PubMed:36638793). Related references: PMID:10676813, PMID:16777602, PMID:24486019, PMID:26245381, PMID:27863242, PMID:36638793
  • Component of the transient SURF complex that recruits UPF1 to stalled ribosomes, linking translation termination to nonsense-mediated mRNA decay (NMD) (PubMed:19417104). Related references: PMID:19417104
  • Required for SHFL-mediated translation termination that suppresses programmed −1 ribosomal frameshifting (−1PRF) on viral and cellular mRNAs (PubMed:30682371). Related references: PMID:30682371
  • Predominantly localized in the cytoplasm.
  • Subject to post-translational modifications including acetylation, methylation, hydroxylation, and phosphorylation.

Experimental Guidance and Technical Tips

  • The immunogen covers amino acids 278–437 in the C-terminal region of human ETF1; verify reactivity against full-length protein and any isoforms in your sample system.
  • For western blotting, a band around 49 kDa is expected; optimize loading and use appropriate positive controls from human, mouse, or rat cell lysates.
  • In immunofluorescence/ICC, expect cytoplasmic staining; include negative controls to confirm specificity.
  • For ELISA, the antibody may be tested as a capture or detection reagent; consider pairing with a suitable matched antibody for sandwich assays.
