Biological Background: ESRRB Function and Localization

• ESRRB, also known as ERR2 or ERR beta, is a nuclear receptor that acts as a transcription factor by binding the canonical ERRE sequence 5'-TCAAGGTCA-3' on target gene promoters and enhancers, thereby regulating gene expression. Related references: PMID:17920186, PMID:19755138

• It plays a critical role in maintaining self-renewal and pluripotency of embryonic and trophoblast stem cells through LIF-independent mechanisms involving FGF and Wnt signaling pathways.

• ESRRB directly activates key pluripotency factors, including GATA6, NR0B1, POU5F1, and PERM1, and co-assembles with SOX2 and POU5F1 on chromatin to drive the embryonic stem cell transcriptional program. Related references: PMID:23836911

• During mitosis, ESRRB remains bound to a subset of its interphase target genes, enabling rapid transcriptional reactivation in early G1 phase to sustain the pluripotency state.

• The immunogen corresponds to the C-terminal region (aa 434–508) of human ESRRB. For Western blot, the predicted molecular weight is approximately 48 kDa; however, alternative splicing or post-translational modifications may give rise to additional bands.
• This rabbit polyclonal antibody has been demonstrated for WB and ELISA using human and mouse samples. When using for other species or applications, consider validating specificity and sensitivity in the relevant sample system.
• As a polyclonal antibody, lot-to-lot variability may occur. It is recommended to titrate the antibody and include appropriate positive and negative controls in each experiment.
