Biological Background: ERAB/HSD17B10 (3-hydroxyacyl-CoA dehydrogenase type-2) Function and Localization

  • Multifunctional mitochondrial enzyme with dual roles: acts as an (S)-3-hydroxyacyl-CoA dehydrogenase in fatty acid beta-oxidation and also catalyzes oxidation of 3-hydroxy-2-methylbutyryl-CoA in isoleucine degradation. Prefers short- and medium-chain substrates with highest efficiency for (3S)-hydroxybutanoyl-CoA. Related references: PMID:10600649, PMID:12917011, PMID:18996107
  • Moonlights as a non-catalytic subunit (MRPP2) of the mitochondrial ribonuclease P complex, involved in tRNA 5'-end cleavage, N1-methylation of guanine and adenine at position 9 in tRNAs, and serving as a tRNA maturation platform to facilitate 3'-processing and CCA addition. Related references: PMID:18984158, PMID:23042678, PMID:24549042, PMID:29040705
  • Exhibits hydroxysteroid dehydrogenase activity, oxidizing 3alpha-, 17beta-, 20beta- and 21-hydroxysteroids as well as 7alpha- and 7beta-hydroxy bile acids. Notably oxidizes allopregnanolone/brexanolone at the 3alpha-hydroxyl group, which may impact GABA-A receptor activation. Related references: PMID:19706438, PMID:28888424
  • Possesses phospholipase C-like activity toward cardiolipin and its oxidized species, likely degrading damaged cardiolipin during oxidative stress to protect cells from apoptosis. Related references: PMID:26338420
  • Interacts with intracellular amyloid-beta, which may contribute to neuronal dysfunction in Alzheimer disease (AD); overexpression observed in AD-affected neurons. Related references: PMID:9338779
  • Subcellular localization: predominantly mitochondrial matrix and associated with mitochondrial nucleoids, essential for mitochondrial structural and functional integrity. Related references: PMID:20077426
  • Post-translational modifications include acetylation, and genetic variants are linked to HSD10 mitochondrial disease (intellectual disability and neurodegeneration).

Experimental Guidance and Technical Tips

  • The immunogen is a synthetic peptide from amino acids 162–261 near the C-terminus. Consider using a blocking peptide to verify specific band detection in WB.
  • Based on its molecular weight (27kDa), ensure adequate gel separation in the 20–35kDa range and transfer conditions optimized for small proteins.
  • WB: validate antibody performance in your target species (human, mouse, rat) using appropriate tissue or cell lysates known to express HSD17B10 (e.g., mitochondria-enriched fractions).
  • ELISA: a direct or indirect ELISA with recombinant antigen or peptide coating may help assess batch-to-batch consistency.
  • For applications involving post-translational modifications or interaction studies, consider that acetylation and amyloid-beta binding may influence epitope accessibility.
