Biological Background: Alpha-enolase (ENO1) Function and Localization
- Alpha-enolase (ENO1) is a multifunctional protein that catalyzes the conversion of 2-phosphoglycerate to phosphoenolpyruvate in glycolysis and the reverse reaction in gluconeogenesis. Related references: PMID:1369209, PMID:29775581
- It is involved in growth control, hypoxia tolerance, and allergic responses. Related references: PMID:10802057, PMID:12666133, PMID:2005901
- On the cell surface, ENO1 functions as a plasminogen receptor and activator, contributing to the fibrinolytic system. Related references: PMID:12666133
- It stimulates immunoglobulin production. Related references: PMID:1369209
- ENO1 also acts as a transcriptional repressor by binding to the myc promoter and may possess tumor suppressor activity.
- Subcellular localization includes the cytoplasm, cell membrane, nucleus, and the M line of sarcomeres in striated muscle.
- The alpha/alpha homodimer is widely expressed in most tissues, whereas tissue-specific heterodimers with beta or gamma enolases are found in muscle and neurons, respectively.
- Post-translational modifications such as acetylation, phosphorylation, and ubiquitination regulate its functions, and it features DNA-binding and transcriptional regulatory activities.
Experimental Guidance and Technical Tips
- For Western blot (WB), use reducing SDS-PAGE with whole-cell lysates from human or mouse cells; a band approximately 47 kDa should be detected.
- In immunohistochemistry (IHC-P), antigen retrieval methods may be required; optimize citrate or EDTA buffer conditions for each tissue type.
- For immunofluorescence (IF/ICC), fixation and permeabilization protocols can affect the detection of ENO1 due to its presence in multiple subcellular compartments; include appropriate controls.
- In immunoprecipitation (IP), the polyclonal nature of this antibody may provide efficient capture of native protein complexes; validate via Western blot.
- For ELISA, this antibody can be used as capture or detection reagent after confirming compatibility with coating buffers and detection systems.
- All applications should include relevant positive and negative control samples; cross-reactivity with mouse allows for use in mouse-derived models.
