Biological Background: ELOVL6 Function and Localization
- ELOVL6 (Very long chain fatty acid elongase 6) is a member of the ELOVL family of fatty acid elongases.
- It catalyzes the first and rate-limiting step in the elongation cycle of long-chain fatty acids, condensing C12, C14, and C16 acyl-CoAs, with highest activity toward C16:0.
- This ER membrane-bound enzyme contributes to the synthesis of unsaturated C16 long chain fatty acids and, to a lesser extent, C18:0 and those with low desaturation degree.
- It may participate in the production of saturated and monounsaturated very long chain fatty acids (VLCFAs) that serve as precursors for membrane lipids and lipid mediators.
- Subcellular localization is at the endoplasmic reticulum membrane.
- Expression is ubiquitous, suggesting a fundamental role in cellular lipid metabolism.
- According to UniProt keywords, ELOVL6 is a glycoprotein, transferase, multi-pass transmembrane protein, and is involved in fatty acid biosynthesis and metabolism.
Experimental Guidance and Technical Tips
- Immunogen design: The antibody was raised against a synthetic peptide spanning amino acids 50–150 of human ELOVL6. This region is predicted to be intracellular, potentially accessible in denaturing conditions for Western blotting.
- Application considerations: For Western blot, consider using denaturing and reducing conditions to expose the epitope. Validate in relevant sample systems expressing ELOVL6, such as human, mouse, or rat tissues/cells.
- For ELISA, the polyclonal nature may provide high sensitivity, but potential cross-reactivity with other ELOVL family members should be assessed.
- Protein size: The predicted molecular weight is 31 kDa, but the observed band may differ due to glycosylation or other post-translational modifications.
