Biological Background: ELAC2 Function and Localization
- ELAC2 is a zinc phosphodiesterase (ElaC homolog protein 2) with multiple aliases including Heredity prostate cancer protein 2 (HPC2), Ribonuclease Z 2 (RNase Z 2), and tRNA 3 endonuclease 2 (tRNase Z 2).
- Functions as a mitochondrial tRNA 3'-processing endonuclease, catalyzing removal of the 3'-trailer from precursor tRNA to facilitate tRNA maturation and ribosome assembly. Related references: PMID:21593607 PMID:24703694
- Localizes primarily to the mitochondrial matrix and nucleoid, with additional nuclear localization.
- Widely expressed across tissues, with high expression in heart, placenta, liver, skeletal muscle, kidney, pancreas, testis, and ovary; lower expression in brain, lung, spleen, thymus, prostate, small intestine, colon, and leukocytes.
- Contains a zinc-binding domain and belongs to the metallo-beta-lactamase superfamily; undergoes alternative splicing and phosphorylation.
- Mutations in ELAC2 are linked to prostate cancer susceptibility (HPC2) and combined oxidative phosphorylation deficiency 17 (COXPD17), highlighting its role in mitochondrial pathology.
Experimental Guidance and Technical Tips
- The immunogen used is a recombinant fusion protein corresponding to amino acids 1-270 of human ELAC2 (NP_776065.1), covering the N-terminal region; this polyclonal antibody may recognize multiple splice variants.
- Recommended for western blot (predicted band around 92 kDa), immunohistochemistry (paraffin-embedded), and ELISA; consider optimizing dilutions and detection conditions for each assay.
- Validated for cross-reactivity with mouse and rat samples; confirm specificity in the relevant biological context.
