Biological Background: EIF4ENIF1 Function and Localization
- EIF4ENIF1 (4E-T) is an eIF4E-binding protein that functions as a transporter and regulator of translation, primarily involved in mRNA storage and decay in processing bodies (P-bodies).
- Acts as a binding platform coordinating deadenylation and decapping repression: it promotes deadenylation via the CCR4-NOT complex and inhibits decapping through eIF4E interaction, thereby protecting deadenylated/repressed mRNAs from degradation. Related references: PMID:27342281 PMID:32354837
- Essential for P-body formation and miRNA-mediated translational repression; it is required for the assembly of P-bodies and facilitates silencing by miRNAs. Related references: PMID:16157702 PMID:24335285 PMID:28487484
- Subcellular localization includes P-bodies, cytoplasm, nucleus, PML bodies, and nuclear speckles, reflecting its dual roles in cytoplasmic mRNA regulation and nuclear transport.
- Mediates nuclear import of EIF4E and DDX6 via a piggy-back mechanism, functioning as a nucleocytoplasmic shuttling protein. Related references: PMID:10856257 PMID:28216671
- Post-translationally modified by acetylation, phosphorylation, isopeptide bond formation, and ubiquitin-like conjugation, which may regulate its localization and activity.
- Widely expressed across tissues, consistent with its fundamental role in mRNA metabolism.
Experimental Guidance and Technical Tips
- The immunogen (residues 150–250) lies in a region that may share conservation across species; consider sequence alignment if using in non-human samples, though cross-reactivity with mouse and rat is indicated.
- For western blotting (WB), a band near 108 kDa is expected; validate the observed molecular weight in your sample system, as post-translational modifications may cause slight shifts.
- In immunofluorescence (IF/ICC), EIF4ENIF1 exhibits punctuate cytoplasmic staining (P-bodies) and nuclear speckles; include appropriate negative controls and consider co-staining with P-body markers.
- For ELISA, optimize coating and detection conditions using the synthetic peptide or recombinant protein as a positive control; cross-reactivity with other species should be verified empirically.
