Biological Background: EIF3H Function and Localization

  • EIF3H (eIF3S3, eIF3-p40, eIF3-gamma) is a subunit of the eukaryotic translation initiation factor 3 (eIF-3) complex, which plays a critical role in the initiation of protein synthesis.
  • The eIF-3 complex associates with the 40S ribosomal subunit and facilitates the recruitment of other initiation factors to form the 43S pre-initiation complex (43S PIC), stimulating mRNA recruitment and scanning for AUG recognition. Related references: PMID:17581632
  • EIF3H is involved in the disassembly and recycling of post-termination ribosomal complexes and prevents premature joining of ribosomal subunits prior to initiation. Related references: PMID:17581632
  • The eIF-3 complex specifically targets and regulates translation of mRNAs involved in cell proliferation, including cell cycling, differentiation, and apoptosis, using different RNA stem-loop binding modes. Related references: PMID:25849773 PMID:27462815
  • EIF3H is localized in the cytoplasm.
  • Post-translational modifications of EIF3H include phosphorylation and isopeptide bond formation.

Experimental Guidance and Technical Tips

  • The immunogen used is a recombinant fusion protein encompassing amino acids 80-340 of human EIF3H, which may contribute to the antibody's specificity.
  • For Western blotting, a band around 40 kDa is expected based on the predicted protein weight; use appropriate controls to confirm specific detection.
  • This antibody is reported to work in WB, IP, IF/ICC, and ELISA; for immunoprecipitation and immunofluorescence, optimization of antibody concentration and fixation conditions is advised.
  • Cross-reactivity is noted for human, mouse, and rat, but we recommend verifying performance in each species and application of interest.
