Biological Background: EHD4 Function and Localization

  • EHD4 (EH domain-containing protein 4), also referred to as PAST4 or hepatocellular carcinoma-associated protein 10/11, is a member of the EHD ATPase family involved in intracellular membrane trafficking.
  • It acts as an ATP- and membrane-binding protein that orchestrates membrane reorganization and tubulation upon ATP hydrolysis, playing a critical role in early endosomal transport (PubMed:17233914, PubMed:18331452). Related references: PMID:17233914 PMID:18331452
  • During sprouting angiogenesis, EHD4 forms a complex with PACSIN2 and MICALL1 to generate recycling endosome-like tubular structures at asymmetric adherens junctions, thereby regulating CDH5 trafficking.
  • Subcellularly, EHD4 is associated with early endosome and recycling endosome membranes, and it also localizes to the cell membrane and adherens junctions.
  • EHD4 is highly expressed in pancreas and heart, indicating tissue-specific functional roles.
  • The protein undergoes acetylation and phosphorylation, and it features ATP-binding and calcium-binding motifs that support its regulatory activities in membrane dynamics.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 362–541 (C-terminal region) of human EHD4; verify that this region is conserved in your target species if using non‑human/mouse samples.
  • For Western blot, a band near 61 kDa is expected; consider using positive control lysates from EHD4‑expressing tissues such as pancreas or heart.
  • In ELISA, use a recombinant EHD4 fragment or full‑length protein as a standard and titrate the antibody to determine optimal coating and detection conditions.
  • As a polyclonal antibody, evaluate lot‑to‑lot consistency for reproducibility in quantitative assays.
