Biological Background: Transcription factor E2F6 Function and Localization
- Transcriptional repressor: E2F6 acts as an inhibitor of E2F-dependent transcription. It binds DNA cooperatively with DP proteins through the E2 recognition site 5'-TTTC[CG]CGC-3', with a preference for 5'-TTTCCCGC-3'. Unlike activator E2Fs, E2F6 lacks transcriptional activation and pocket protein binding domains. Related references: PMID:9501179, PMID:9704927
- Cell cycle regulation: E2F6 regulates a subset of E2F-dependent genes whose products are required for entry into the cell cycle but not for normal cell cycle progression. Overexpression delays the exit of cells from S-phase. Related references: PMID:9501179, PMID:9689056
- Repression mechanism: It may silence gene expression by recruiting a chromatin remodeling complex that contains histone H3-K9 methyltransferase activity.
- Meiosis-specific gene repression: E2F6 represses expression of some meiosis-specific genes, including SLC25A31/ANT4 (by similarity).

- Predicted molecular weight is 32 kDa; however, endogenous E2F6 may appear at slightly different sizes due to post-translational modifications. Always validate the band pattern in your sample system.
- For ChIP applications, while the antibody is raised against a recombinant protein, we recommend performing a ChIP-grade validation in your specific cell type and target locus.
