Biological Background: E2F1 Function and Localization
- E2F1 (Transcription factor E2F1), also designated PBR3, RBAP-1, or RBBP-3, is a member of the E2F family of transcription factors.
- E2F1 binds DNA cooperatively with DP proteins through the E2 recognition site (5'-TTTC[CG]CGC-3') found in promoters of genes involved in cell cycle regulation and DNA replication. The DRTF1/E2F complex controls G1-to-S phase transition, and E2F1 binds RB1 in a cell-cycle dependent manner. Related references: PMID:10675335 PMID:12717439 PMID:17050006 PMID:17704056
- It can mediate both cell proliferation and TP53/p53-dependent apoptosis. Related references: PMID:8170954
- Blocks adipocyte differentiation by binding to specific promoters and repressing CEBPA binding to its target gene promoters. Related references: PMID:20176812
- Directly activates transcription of PEG10, and positively regulates transcription of RRP1B. Related references: PMID:17050006 PMID:18625225 PMID:28992046 PMID:20040599
- Subcellular localization: Nucleus.
- Post-translational modifications include acetylation, phosphorylation, methylation, and ubiquitination; also involved in host-virus interactions.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the C-terminal region (aa 301-437) of human E2F1; consider its suitability for detecting full-length E2F1 or specific isoforms.
- For Western blotting, validate detection in nuclear extracts from human, mouse, or rat samples, given the antibody's cross-reactivity.
- For ELISA, the polyclonal nature may be advantageous for capture or detection in sandwich assays; optimize coating concentrations and incubation conditions.
- Since E2F1 is a transcription factor predominantly localized in the nucleus, enrich nuclear fractions to improve signal intensity.
