Biological Background: DUT Function and Localization
- Deoxyuridine 5'-triphosphate nucleotidohydrolase, mitochondrial (dUTP pyrophosphatase), also known as DUT, is a key enzyme in nucleotide metabolism encoded by the DUT gene.
- It catalyzes the cleavage of dUTP into dUMP and inorganic pyrophosphate, thereby preventing uracil misincorporation into DNA and supplying the substrate (dUMP) for de novo thymidylate biosynthesis. Related references: PMID:17880943 PMID:8631816 PMID:8805593
- Additionally, the N-terminal domain of DUT can inhibit peroxisome proliferator-activated receptor (PPAR) activity by binding to PPAR and preventing its dimerization with retinoid X receptor, though this function is inferred by similarity.
- DUT localizes to both the nucleus and the mitochondrion, consistent with its dual role in nuclear DNA replication and mitochondrial metabolism.
- Tissue-specific expression: isoform 3 is broadly expressed constitutively, while isoform 2 expression correlates with the onset of DNA replication and its degradation coincides with the cessation of nuclear DNA replication.
- The canonical protein has a calculated molecular weight of approximately 27 kDa (UniProt: 26563 Da) and undergoes phosphorylation; it is associated with keywords including magnesium binding, hydrolase activity, and alternative splicing.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fusion protein spanning amino acids 160 to 252 of human DUT (isoform 1, NP_001020419.1), which corresponds to the C-terminal region. This region may be conserved across DUT isoforms; therefore, the antibody may detect multiple isoforms, but isoform-specific reactivity should be verified experimentally.
- For Western blot (WB), the predicted molecular weight is 27 kDa. Use appropriate positive controls (e.g., human cell lines) and validate the band pattern under your experimental conditions.
- This rabbit monoclonal antibody is suitable for ELISA. Consider optimizing coating antigen concentration and antibody dilution for your specific assay format.
- Shipping condition: ice bag; upon receipt, store at recommended temperature to maintain stability.
