Biological Background: DNMT3A Function and Localization
- DNMT3A is required for genome-wide de novo DNA methylation and is essential for establishing DNA methylation patterns during development. It modifies DNA in a non-processive manner and also methylates non-CpG sites. DNA methylation is coordinated with methylation of histones. - May preferentially methylate DNA linker between two nucleosomal cores and is inhibited by histone H1 (By similarity).
- Plays a role in paternal and maternal imprinting and is required for methylation of most imprinted loci in germ cells (By similarity).
- Acts as a transcriptional corepressor for ZBTB18, recruits HDAC activity, and is recruited to trimethylated 'Lys-36' of histone H3 (H3K36me3) sites (By similarity).
- Exhibits weak auto-methylation activity on Cys-710 in the absence of DNA (By similarity).
- Subcellular localization: predominantly nuclear, associated with chromosomes, and also detected in the cytoplasm.
- Tissue expression: highly expressed in fetal tissues, skeletal muscle, heart, peripheral blood mononuclear cells, and kidney; lower expression in placenta, brain, liver, colon, spleen, small intestine, and lung.
- Alternative promoter usage and alternative splicing generate multiple isoforms, including the shorter DNMT3A2. The protein contains zinc-finger domains, a methyltransferase domain, and is subject to phosphorylation and ubiquitination.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the N-terminal region (amino acids 1–132) of human DNMT3A. Consider that isoforms lacking this region (e.g., certain splice variants) may not be detected.
- For Western blot, the predicted molecular weight of the full-length protein is ~102 kDa. Validate the observed bands using positive controls or recombinant protein; be aware of potential isoform variations.
- For immunoprecipitation, optimize the antibody concentration and incubation conditions with your specific sample type. Nuclear extraction protocols are recommended due to the predominant nuclear localization of DNMT3A.
- The antibody shows cross-reactivity with mouse and rat DNMT3A. When targeting other species, verify sequence homology in the immunogen region.
