Biological Background: eEF1A Function and Localization

  • eEF1A (eukaryotic elongation factor 1A) exists as two isoforms, eEF1A1 (gene EEF1A1, GeneID: 1915) and eEF1A2 (gene EEF1A2, GeneID: 1917), both highly conserved translation elongation factors. They catalyze the GTP-dependent binding of aminoacyl-tRNA to the ribosomal A-site during protein synthesis elongation PMID:26593721, PMID:26651998, PMID:36123449, PMID:36264623, PMID:36638793.
  • Beyond translation, eEF1A1 promotes actin bundling for cytoskeleton organization and, in T-helper 1 cells, forms a complex with TXK and PARP1 to positively regulate IFNG transcription PMID:17177976. EEF1A1 is also required for the translation of viral proteins during SARS-CoV-2 infection PMID:33495306.
  • EEF1A1 is broadly expressed, while EEF1A2 is found predominantly in brain, heart, and skeletal muscle. Subcellularly, EEF1A1 localizes to the cytoplasm, nucleus, nucleolus, and cell membrane; EEF1A2 localizes to the endoplasmic reticulum membrane PMID:19158340, PMID:8650580, PMID:26497934, PMID:36697828.
  • Lysine 55 (K55) is a conserved dimethylation site (N6,N6-dimethyllysine) in both eEF1A1 and eEF1A2, as identified by proteomics PMID:30143613, PMID:30612740. In eEF1A2, K55 can also be trimethylated. These modifications may regulate protein interactions and translation fidelity.
  • Mutations in EEF1A2 are linked to neurodevelopmental disorders such as epilepsy and intellectual disability, underscoring the biomedical importance of eEF1A2 regulation.

Experimental Guidance and Technical Tips

  • This antibody was generated against a dimethylated K55 peptide from eEF1A; it should specifically recognize the dimethyl-K55 form. Confirm specificity by peptide competition (dimethyl vs. unmodified) or by treating samples with a demethylase.
  • For WB, eEF1A1 (~50.1 kDa) and eEF1A2 (~50.5 kDa) migrate at ~50 kDa. Use standard SDS-PAGE and transfer conditions. Validate in human cell lines (e.g., HeLa for EEF1A1, SH-SY5Y for EEF1A2).
  • In ELISA, coat with dimethylated peptide or cell lysate; include non-methylated controls to assess cross-reactivity.
  • Given the role of eEF1A in translation, cancer biology, and viral replication, this antibody can aid studies of methylation-dependent regulation under various physiological and pathological conditions.
