Biological Background: DGCR8 Function and Localization

  • DGCR8, also known as DiGeorge syndrome critical region 8 or pasha (Gene symbol: C22orf12, DGCRK6), is a subunit of the microprocessor complex.
  • Functions as a molecular anchor within the microprocessor complex, recognizing pri-miRNA at dsRNA-ssRNA junctions and directing DROSHA to cleave 11 bp away, producing pre-miRNA for further processing by DICER. Related references: PMID:26027739 PMID:26748718
  • Its activity is heme-dependent: the heme-bound DGCR8 dimer binds pri-miRNAs cooperatively as a trimer of dimers to trigger cleavage, whereas the heme-free monomer is much less active. Related references: PMID:15531877 PMID:15574589 PMID:15589161
  • Specifically recognizes N6-methyladenosine (m6A)-modified pri-miRNAs, a modification critical for efficient pri-miRNA processing. Related references: PMID:25799998
