Biological Background: DDX50 Function and Localization

  • DDX50 (DEAD box protein 50), also known as Gu-beta or nucleolar protein Gu2, is an ATP-dependent RNA helicase involved in pre-mRNA splicing and ribosomal RNA production. Related references: PMID:12027455

  • DDX50 acts as a viral restriction factor, inhibiting replication of vaccinia virus, herpes simplex virus, Zika virus, and dengue virus during early infection. It activates NF-kappa-B and IRF3 signaling through the adapter TICAM1, leading to interferon-beta production. Related references: PMID:28181036, PMID:35215908

  • The protein localizes to the nucleolus, nucleus, and cytoplasm.

  • Highest expression is observed in skeletal muscle, liver, heart, placenta, and kidney.

  • DDX50 contains conserved helicase ATP-binding and RNA-binding domains and is a phosphoprotein subject to ubiquitin-like conjugation.

Experimental Guidance and Technical Tips

  • The immunogen covers amino acids 1–135 (N-terminal region) of human DDX50; confirm reactivity with full-length target in your experimental system.
  • For Western blotting, a band near 83 kDa is expected; use appropriate molecular weight markers.
  • The antibody is reactive with human, mouse, and rat samples, supporting multi-species applications.
  • Given nucleolar localization, immunofluorescence may show nucleolar enrichment; include DAPI or nucleolar marker co-staining.
  • In antiviral studies, evaluate activity in relevant cell lines and consider viral infection time courses.
  • Standard ELISA and IHC-P protocols can be adapted; optimize dilution and detection conditions empirically.
