Biological Background: DDX1 Function and Localization

  • ATP-dependent RNA helicase able to unwind RNA-RNA and RNA-DNA duplexes; possesses ATPase activity on RNA.
  • Acts as a coactivator with RELA to enhance NF-kappa-B-mediated transcription; binds cyclin CCND2 promoter.
  • Binds poly(A) RNA and may be involved in mRNA 3'-end processing and polyadenylation.
  • Accessory subunit of the tRNA-splicing ligase complex, directly joining spliced tRNA halves; cooperates with archease for RTCB guanylylation. Related references: PMID:24870230
  • Subcellularly located in nucleus, cytoplasm, cytoplasmic granules, cytosol, and mitochondrion.
  • Highest expression in tissues of neuroectodermal origin (retina, brain, spinal cord) and retinoblastoma cell lines.
  • Component of a cytoplasmic sensor for viral dsRNA, triggering antiviral innate immune responses via TICAM1.

  • The immunogen spans amino acids 441–740 within the C-terminal helicase domain, which is conserved across human, mouse, and rat, supporting the observed cross-reactivity.
  • For Western blot, a band near 82 kDa is expected; consider using lysates from human, mouse, or rat cell lines as positive controls.
  • IHC-P applications may require antigen retrieval optimization; retinoblastoma or brain tissue sections are recommended for validation.
  • In immunoprecipitation, due to DDX1’s multiple subcellular locations, lysis buffer composition and salt concentration may need adjustment.
  • ELISA performance should be verified with recombinant DDX1 protein or endogenous detection in cell extracts.
