Biological Background: DAB2IP Function and Localization
- DAB2IP, also known as ASK-interacting protein 1 (AIP-1) or DOC-2/DAB-2 interactive protein, is encoded by the DAB2IP gene. Multiple isoforms are produced by alternative splicing.
- Functions as a scaffold protein that regulates a broad spectrum of signaling pathways, including innate immune response, inflammation, cell proliferation, apoptosis, angiogenesis, and cell migration.
- Acts as a GTPase-activating protein (GAP) for ARF6, Ras, and RAB40C, promoting GTP hydrolysis and negatively regulating pathways such as TLR4-dependent NF-kappa-B signaling.

PMID:29156729 - Mediates TNF-induced apoptosis by facilitating 14-3-3 dissociation from MAP3K5, activating the JNK signaling pathway, while also inhibiting NF-kappa-B signaling.
- Regulates the balance between PI3K-AKT survival and MAP3K5-JNK stress signaling, modulating phosphorylation status of AKT1 and MAP3K5 in response to pro-inflammatory stimuli.
- Located in the cytoplasm, cell membrane, and dendrites; expressed in endothelial cells, vascular smooth muscle cells, and prostate epithelial cells, with down-regulation in prostate cancer.
- Plays a role in cell cycle checkpoint control by reducing G1 phase cyclin levels, leading to G0/G1 arrest, and acts as a tumor suppressor in prostate cancer by preventing EMT and inhibiting PI3K-AKT and Ras-MAPK pathways.
- Involved in neuronal development, promoting cortical neuron radial migration and Purkinje cell dendrite development as a downstream effector of the Reelin pathway.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 205-320 of human DAB2IP, a region that may be involved in protein interactions. Consider sequence alignment to assess conservation across species if cross-reactivity is expected beyond human and rat.
- For Western blotting, DAB2IP has a predicted molecular weight of ~132 kDa; optimize protein extraction and electrophoresis conditions to resolve high-molecular-weight bands, and test antibody dilutions empirically.
- In immunofluorescence/ICC, DAB2IP localizes to cytoplasm, plasma membrane, and cell projections; include appropriate fixation and permeabilization steps, and co-staining with organelle markers to confirm subcellular distribution.
- ELISA performance may require optimization of coating antigen and antibody concentration; use recombinant DAB2IP protein or relevant biological samples as positive controls.
