  • Cytokeratin 20 (CK20), also designated Keratin-20 or Protein IT, is a type I cytoskeletal keratin encoded by the KRT20 gene.
  • Plays a significant role in maintaining keratin filament organization in intestinal epithelia; upon phosphorylation, it contributes to mucin secretion in the small intestine (By similarity).
  • As a member of the intermediate filament family, CK20 contains a coiled-coil domain and is annotated with keywords including apoptosis, phosphoprotein, and direct protein sequencing.
  • Predominantly expressed in the intestinal epithelium (luminal cells of colonic mucosa) and in Merkel cells of the oral cavity (gingival, palatal, and lingual mucosa).
  • Subcellular localization is cytoplasmic, consistent with its structural role.
  • Due to its tissue-restricted expression pattern, CK20 is widely used as an immunohistochemical marker for gastrointestinal epithelium and Merkel cell-derived tumors.

  • The immunogen is a synthetic peptide mapping to the C‑terminal region (aa 325–424) of human CK20, a domain that is unique and well‑suited for generating specific antibodies.
  • For Western blotting, a band at ~48 kDa is expected; consider using intestinal or colon tissue lysates as a positive control.
  • For IHC‑P, heat‑induced epitope retrieval (e.g., citrate buffer, pH 6.0) may improve detection; validate expression in normal intestinal or Merkel cell‑containing tissues and use isotype controls.
  • It is recommended to titrate the antibody for each application and include both positive and negative control lysates/tissues.
