Biological Background: Cytokeratin 14 (KRT14) Function and Localization
- Keratin 14 (KRT14) is a member of the type I keratin family, also known as cytokeratin-14 (CK-14) or keratin-14 (K14), and serves as a structural protein in epithelial cells.
- It forms obligate heteropolymers with the type II keratin KRT5, contributing to the keratin intermediate filament network that provides mechanical strength and structural integrity to the basal keratinocytes of stratified epithelia (PubMed:11724817, PubMed:1694855, PubMed:1720261). - The nonhelical tail domain of KRT14 promotes KRT5-KRT14 filaments to self-organize into large bundles, enhancing the mechanical resilience of keratin intermediate filaments in vitro (PubMed:11724817). Related references: PMID:11724817
- KRT14 is predominantly localized to the cytoplasm, but can also be found in the nucleus.
- It is expressed in the corneal epithelium and is detected in the basal layer of the epidermis, with reduced expression in the more apically located layers (stratum spinosum, stratum granulosum) and absence in the stratum corneum. It is also strongly expressed in the outer root sheath of anagen hair follicles.
- Post-translational modifications include phosphorylation and ubiquitination, and mutations in KRT14 are associated with several skin fragility disorders such as epidermolysis bullosa simplex and ectodermal dysplasia.
